B. Riley Says EyePoint Phase 3 Miss Strengthens 4D Molecular’s Investment Case

B. Riley said EyePoint Pharmaceuticals’ failed Phase 3 study for Duravyu may ultimately strengthen the investment case for 4D Molecular Therapeutics (NASDAQ: FDMT), particularly from both competitive and mechanistic perspectives.

B. Riley analyst Mayank Mamtani reiterated a Buy rating on 4D Molecular with a $37 price target after EyePoint Pharmaceuticals (NASDAQ: EYPT) reported that its Phase 3 trial of Duravyu in wet age-related macular degeneration did not meet its primary endpoint.

Shares of 4D Molecular rose about 12% to $13.70 in afternoon trading following the development.

EyePoint Study Miss Seen as Competitive Positive for 4D-150

According to B. Riley, the Duravyu setback reduces competitive pressure around 4D Molecular’s lead ophthalmology program, 4D-150.

Mamtani argued that the EyePoint result should be viewed as a net positive for 4D-150 because the two programs differ meaningfully in both trial design and mechanism.

A key distinction, according to the firm, lies in patient selection.

B. Riley said 4D-150’s Phase 3 entry criteria exclude seven of the nine etiologies that contributed to what it described as a confounded cohort in EyePoint’s LUGANO study.

That could make 4D-150’s pivotal dataset easier to interpret and potentially reduce the risk that unrelated disease factors obscure treatment efficacy.

B. Riley Highlights Mechanistic Advantage of 4D-150

The firm also emphasized the underlying biology of 4D-150.

4D-150 is designed to express aflibercept, an established anti-VEGF therapy already widely used in retinal disease.

Because aflibercept itself is also used in 4D-150’s control arm, B. Riley believes the program benefits from what it described as an established relationship between retinal anatomy and visual outcomes for the same molecule.

The analyst characterized this as a meaningful mechanistic distinction that could influence trial outcomes in addition to execution quality.

In other words, 4D-150 is not relying on an entirely new therapeutic mechanism to establish efficacy. Instead, it is seeking to deliver sustained expression of a molecule whose clinical effects are already well characterized.

4D Molecular Shares Rise After EyePoint Setback

Investors appeared to react positively to the competitive implications of EyePoint’s failed study.

4D Molecular shares were up approximately 12% to $13.70 in afternoon trading, while B. Riley maintained its $37 price target.

The firm’s thesis is that the EyePoint result potentially improves the competitive positioning of 4D-150 while also reinforcing confidence in the design of 4D Molecular’s Phase 3 program.

The broader read-through is that setbacks among rival wet AMD programs could increase investor focus on companies with differentiated delivery systems, cleaner trial populations and mechanisms anchored to established anti-VEGF therapies.
