RASONQUE Lung Cancer Data Show 42% Response Rate

RASONQUE lung cancer data published in the New England Journal of Medicine showed antitumor activity in previously treated patients with advanced RAS-mutant non-small cell lung cancer.

Revolution Medicines (Nasdaq: RVMD) said on its official press-release page that the Phase 1/2 study evaluated once-daily doses of RASONQUE, also known as daraxonrasib, in 136 patients whose tumors carried RAS mutations other than RAS G12C.

In a selected subgroup of 38 docetaxel-naive patients treated at doses of 160 milligrams to 220 milligrams, the confirmed objective response rate was 42% and the disease-control rate was 89%. Median progression-free survival was 8.3 months and median overall survival was 16 months.

RASONQUE Lung Cancer Data Support a Phase 3 Test

Grade 3 treatment-related adverse events occurred in 25% of patients in the dose group, most commonly rash and diarrhea. The company reported no Grade 4 or Grade 5 treatment-related adverse events at those dose levels.

The findings informed the ongoing RASolve 301 trial, a randomized Phase 3 study comparing RASONQUE with docetaxel in previously treated locally advanced or metastatic RAS-mutant lung cancer. RASONQUE remains investigational for this lung cancer indication despite its separate U.S. approval in metastatic pancreatic cancer.

The response figures come from an early-stage, nonrandomized subgroup and should not be interpreted as proof of benefit over standard treatment. The Phase 3 trial will be needed to test whether the activity translates into a clinically meaningful advantage in a larger controlled population.
